A classical protocol for producing enzymes (CV-wTa) by bacterial cell culture (E. Coli) has been translated on OnePot. The process consists of the following steps: LB medium preparation and sterilisation, E. Coli pre-culture, E. Coli central culture, induction of enzyme expression by IPTG solution, and harvesting. Cell culture yield has been measured by the cellular density (ODD) ratio. The enzyme expression was evaluated by SDS-page analysis.

Monastrol has been synthesised by Biginelli reaction following an MW-assisted protocol previously reported by Kappe et al. The multicomponent reaction between thiourea, 3-OH-benzaldehyde, and ethylacetoacetate is catalysed by Yt(OTf)3 under MW heating at 120°C for 30 min. Compared to classical protocols, MW heating is an instant on-off internal heating system that reduces reaction time and byproducts. Matrix-in-batch technology, implemented by OnePot, is comparable to MW in terms of efficiency on a bigger scale.

Boc protection of amines is a classical procedure of organic chemistry to mask amine group
reactivity. The classical protocol, reacting the amino acid tyrosine in a basic solution by
NaOH at room temperature, followed by HCl neutralization and liquid/liquid extraction, has
been fully reproduced in an automated way.

Zein is a prolamin derived from corn that was proposed for several pharmaceutical and food applications due to its safety profile and low
immunogenicity. Zein-based nanoparticles were prepared by means of the nanoprecipitation technique. In detail, the traditional method was based
on the use of an initial volume of 8 ml of solvent (containing 10 mg of protein), while the OnePot®-assisted process was performed on 320 ml of
formulation (corresponding to ~40 cumulative traditional batches, 400 mg of protein).
Synthesis of PVP K-90 polymer
Povidone (polivnyl pyrrolidone, PVP) polimer has been synthetized following a procedure for the radical polymerization of 1-vinyl-
2-pyrrolidone (VP) activated by 2 2'-azobis(2-methyl)propionitrile (AzBn) as radical initiator. Reaction was controlled by relative
viscosity measure of a 10% water solution of product. Depending on reaction time and temperature it is possible to obtain
different degree of polymerization for different type of PVP, characterized by different K-values, related to relative viscosity in
water. European Pharmacopeia admits PVP with K-value ranging between K-12 and K-90.

CommitmentTo end an existing contract, contact us at least 20 days before the next billing period. Once you have notified us via email or a phone call, we’ll help you book a date to return your OnePot. You can of course keep using your OnePot until the return date.

Maintenance & ServiceWith OnePot's connected features, there's no need to keep track of your maintenance schedule. When it's time for maintenance, we will contact you to schedule a day and time that's convenient for you. Then we'll come to your lab, fresh OnePot up, and handle it back to you. In case anything brakes up, call us and in 48 hours we will be in your lab to fix it.
